Hay genes <\/b>que no solo definen c\u00f3mo se vive, sino tambi\u00e9n cu\u00e1nto tiempo se vive bien<\/b>. <\/p>\n

El alelo APOE2 <\/b>del gen apolipoprote\u00edna E<\/b> es uno de ellos: protege las neuronas <\/b>del da\u00f1o en el ADN <\/b><\/span>y frena el envejecimiento celular del cerebro.<\/p>\n

Las personas con esta variante tienen menos probabilidades de desarrollar la enfermedad de Alzheimer <\/b><\/span>y muestran se\u00f1ales de un envejecimiento cerebral m\u00e1s saludable que quienes portan APOE3 <\/b>o APOE4<\/b>.<\/p>\n

La investigaci\u00f3n fue realizada por el cient\u00edfico mexicano Cristian Ger\u00f3nimo-Olvera<\/b> junto con Lisa Ellerby y un equipo del Instituto Buck para la Investigaci\u00f3n del Envejecimiento, en California<\/b>, en colaboraci\u00f3n con investigadores de la Universidad del Sur de California y la Universidad de Washington, en Estados Unidos. Los resultados se publicaron en la revista Aging Cell<\/b><\/i><\/span>.<\/p>\n

El gen que puede frenar el Alzheimer<\/h2>\n

\"Las<\/p>\n

Durante d\u00e9cadas, los cient\u00edficos sab\u00edan que APOE2 <\/b>se asocia a una vida m\u00e1s larga y a menor riesgo de Alzheimer, pero nadie hab\u00eda explicado por qu\u00e9 a nivel celular. La pregunta era concreta: \u00bfqu\u00e9 hace esta variante dentro de las neuronas que las otras no hacen?<\/b><\/p>\n

Las neuronas son c\u00e9lulas que no se dividen, lo que las hace especialmente vulnerables al da\u00f1o acumulado con los a\u00f1os<\/b>. Cuando ese da\u00f1o se acumula sin repararse, las c\u00e9lulas entran en senescencia, un estado de deterioro que alimenta enfermedades como el Alzheimer.<\/p>\n

La variante APOE4 es el factor de riesgo gen\u00e9tico m\u00e1s fuerte para el Alzheimer. Con una sola copia, el riesgo se cuadruplica; con dos, se multiplica por catorce frente a la variante m\u00e1s com\u00fan, APOE3.<\/p>\n

Los investigadores que publicaron el estudio en Aging Cell<\/b><\/i> quisieron comparar directamente los efectos de APOE2, APOE3 y APOE4 en neuronas humanas para entender qu\u00e9 mecanismos explican las diferencias en longevidad y vulnerabilidad al Alzheimer.<\/p>\n

Neuronas bajo la lupa<\/h2>\n

\"Las<\/p>\n

Los investigadores usaron c\u00e9lulas madre pluripotentes inducidas, es decir, c\u00e9lulas reprogramadas en laboratorio para convertirse en neuronas con cada variante del gen APOE. Trabajaron con dos tipos: las GABA\u00e9rgicas, que frenan la actividad cerebral, y las glutamat\u00e9rgicas, que la activan.<\/p>\n

Para medir la resistencia de las c\u00e9lulas, aplicaron radiaci\u00f3n y doxorrubicina, un f\u00e1rmaco que da\u00f1a el ADN. Luego midieron marcadores de senescencia celular como p16 y CRYAB, dos prote\u00ednas que funcionan como se\u00f1ales de alarma del envejecimiento celular.<\/p>\n

Las neuronas con APOE2 mostraron niveles m\u00e1s bajos de esas prote\u00ednas de alarma y una estructura nuclear mejor conservada que las neuronas con APOE4, tanto en condiciones normales como bajo estr\u00e9s.<\/p>\n

\"Experimentos<\/p>\n

El an\u00e1lisis gen\u00e9tico de c\u00e9lula por c\u00e9lula revel\u00f3 que las neuronas APOE2 activan m\u00e1s genes de reparaci\u00f3n del ADN, entre ellos BRCA1, CDK1, PLK1 y TOP2A, genes que coordinan la detecci\u00f3n y correcci\u00f3n de roturas en el material gen\u00e9tico.<\/p>\n

Los experimentos con ratones de 16 meses confirmaron los resultados: los animales con APOE2 conservaron mejor la prote\u00edna Lamin A\/C en el giro dentado del hipocampo, una regi\u00f3n del cerebro vinculada a la memoria y al Alzheimer. Esta prote\u00edna mantiene el n\u00facleo celular estable y su p\u00e9rdida es una se\u00f1al cl\u00e1sica de envejecimiento.<\/p>\n

Un dato que abri\u00f3 nuevas posibilidades fue que al agregar prote\u00edna APOE2 de laboratorio a neuronas con APOE4, el da\u00f1o al ADN se redujo. Esto sugiere que la protecci\u00f3n no es exclusiva de quienes nacen con esa variante.<\/p>\n

Lo que falta saber y lo que ya cambi\u00f3<\/h2>\n

\"El<\/p>\n

Los investigadores advirtieron que a\u00fan se desconoce el mecanismo molecular exacto por el que APOE2 estabiliza la envoltura nuclear y protege el material gen\u00e9tico. Futuros estudios deber\u00e1n responder esa pregunta antes de trasladar estos hallazgos a tratamientos concretos.<\/p>\n

El estudio abre la puerta a terapias que imiten el efecto de APOE2 en el cerebro, con foco en personas con APOE4, el grupo con mayor riesgo gen\u00e9tico de Alzheimer. <\/p>\n

Esta investigaci\u00f3n estableci\u00f3 que APOE2 va mucho m\u00e1s all\u00e1 del transporte de colesterol: tambi\u00e9n protege el ADN neuronal y podr\u00eda ser clave para entender por qu\u00e9 algunas personas envejecen con el cerebro intacto.<\/p>\n","protected":false},"excerpt":{"rendered":"
