Un grupo de investigadores en la India <\/b>present\u00f3 un modelo molecular innovador que ofreci\u00f3 una explicaci\u00f3n alternativa sobre la formaci\u00f3n de los cuerpos de Lewy<\/b>, estructuras caracter\u00edsticas de la enfermedad de <\/b>Parkinson<\/b><\/span> y otras neuropat\u00edas. <\/p>\n

Seg\u00fan los datos publicados<\/span> en Communications Biology<\/i>, estos dep\u00f3sitos cerebrales se forman porque determinadas variantes de la prote\u00edna \u03b1-sinucle\u00edna<\/b> son capaces de atrapar, de manera indiscriminada, prote\u00ednas y org\u00e1nulos celulares. Este hallazgo permite imaginar una nueva direcci\u00f3n para el desarrollo de futuros tratamientos dirigidos a frenar la neurodegeneraci\u00f3n.<\/p>\n

\u00bfQu\u00e9 son los cuerpos de Lewy y cu\u00e1l es su relevancia<\/h2>\n

Los cuerpos de Lewy son acumulaciones an\u00f3malas de prote\u00ednas <\/b>detectadas en las neuronas de personas que sufren enfermedades neurodegenerativas, en especial el Parkinson. <\/p>\n

Estas inclusiones aparecen en regiones clave del sistema nervioso y se relacionan con s\u00edntomas motores, como temblores y rigidez,<\/b> adem\u00e1s de manifestaciones no motoras, como trastornos del sue\u00f1o<\/b> y alteraciones auton\u00f3micas. <\/p>\n

\"El<\/p>\n

Durante a\u00f1os, la formaci\u00f3n de los cuerpos de Lewy se consider\u00f3 uno de los grandes enigmas de la neurolog\u00eda<\/b>, ya que se sab\u00eda que estaban presentes en la mayor\u00eda de los casos de Parkinson, pero no se comprend\u00eda plenamente c\u00f3mo ni por qu\u00e9 se desarrollaban.<\/p>\n

La \u03b1-sinucle\u00edna<\/b> es una prote\u00edna abundante en las neuronas. En condiciones normales, presenta una estructura desordenada pero estable, con un n\u00facleo hidrof\u00f3bico protegido por interacciones internas que la mantienen en estado inerte. <\/p>\n

El estudio dice que ciertas variantes de la prote\u00edna \u03b1-sinucle\u00edna, relacionadas con la enfermedad, cambian su forma<\/b>. En estas variantes, una parte interna de la prote\u00edna que normalmente est\u00e1 escondida (el n\u00facleo hidrof\u00f3bico) queda al descubierto.<\/b> Esto hace que la prote\u00edna sea m\u00e1s activa<\/b> y act\u00fae como una especie de \u201ctrampa\u201d<\/b> que atrapa otras prote\u00ednas y partes de la c\u00e9lula sin control.<\/p>\n

M\u00e1s all\u00e1 de las teor\u00edas tradicionales<\/h2>\n

Durante mucho tiempo, los modelos cl\u00e1sicos explicaban la formaci\u00f3n de cuerpos de Lewy como resultado de la agregaci\u00f3n fibrilar de la \u03b1-sinucle\u00edna o su separaci\u00f3n en fases l\u00edquidas. <\/p>\n

\"El<\/p>\n

Sin embargo, estas teor\u00edas no lograban justificar por qu\u00e9 los cuerpos de Lewy contienen cientos de componentes distintos<\/b>, como l\u00edpidos y org\u00e1nulos da\u00f1ados, ni c\u00f3mo las variantes alteradas de la prote\u00edna generan la acumulaci\u00f3n y el crecimiento de estas inclusiones. <\/p>\n

El Modelo Multifactorial de Desorden Aleatorio<\/b>, propuesto por cient\u00edficos de India, dice que no solo importa que la prote\u00edna \u03b1-sinucle\u00edna se agrupe, como se pensaba antes. Seg\u00fan este modelo, cuando una parte interna de la prote\u00edna queda expuesta, esto provoca que la prote\u00edna forme conexiones incorrectas y desordenadas con muchas otras prote\u00ednas y partes de la c\u00e9lula.<\/p>\n

El equipo dirigido por Sneha Jos<\/b> y Sivaraman Padavattan<\/b> demostr\u00f3 experimentalmente que las variantes patol\u00f3gicas de la \u03b1-sinucle\u00edna, sobre todo las truncadas, interact\u00faan con una gama mucho m\u00e1s amplia de prote\u00ednas celulares que la prote\u00edna en su forma normal. <\/p>\n

Pruebas de afinidad con diferentes prote\u00ednas, incluidas histonas, BSA, SOD1 y Tau, confirmaron que esta captaci\u00f3n indiscriminada es exclusiva de las variantes asociadas a la enfermedad. Los autores subrayaron en Communications Biology<\/i>: \u201cLas variantes patol\u00f3gicas de \u03b1-sinucle\u00edna act\u00faan como un secuestro molecular,<\/b> atrapando indiscriminadamente prote\u00ednas del entorno celular y formando el n\u00facleo denso de un cuerpo de Lewy\u201d. <\/p>\n
